Recently, Professor Huang He's team from the school of food and pharmaceutical engineering, Nanjing Normal University published a paper entitled "chemical synthesis of glycosaminoglycans" on the top journal accounts of Chemical Research (2019 influencing factor 21.661) in the form of cover (account. Chem. Res. 2020, 53, 335? 346), the research progress of glycosaminoglycans with important physiological activity constructed by chemical enzyme method was reviewed. The corresponding author of this paper is Professor Huang He and Professor Robert J. linhardt of the Department of chemical biology of Rensselaer Institute of technology in the United States. The first author is Dr. Zhang Xing of our university.
Glycosaminoglycans are a kind of polydispersive mucopolysaccharides with complex structure in cells, and are widely used in the development of clinical drugs (such as anticoagulant, anti-inflammatory, etc.). However, there are still some problems in the development of glycosaminoglycans. The strategy of chemical enzyme method is to combine chemical synthesis and enzyme catalysis to simulate the biosynthesis of glycosaminoglycan and realize biomimetic synthesis. This method has the flexibility of chemical synthesis and the high efficiency of enzyme catalysis. The reaction process does not need to protect the group and is regioselective and stereoselective. It is helpful for the research of glycosaminoglycan biology to be precise, controllable and functional.
In 2009 and 2014, Dr. Zhang obtained his bachelor's and doctor's degrees from Beijing Normal University and the University of Chinese Academy of Sciences. Later, he worked as a postdoctoral researcher at Rensselaer Institute of technology in the United States and a research scientist at Albany molecular pharmaceutical company in the United States. Since he joined Nanjing Normal University, Dr. Zhang Xing has made more in-depth work in the field of sugar medicine research, and has published relevant research results in a number of internationally renowned journals. In particular, great breakthroughs have been made in the study of cognitive degradation in patients with sepsis. It is the first time to reveal the close relationship between the fragmentation of endogenous heparin sulfate (one of the main glycosaminoglycans) and cognitive impairment, providing a new target for the treatment of cognitive degradation in patients with sepsis. This work was published in PNAS (PNAs, 2019, 116, 9208-9213).
In recent years, Professor Huang He's team has carried out a series of in-depth studies on the design and synthesis of active natural products based on microbial resources, combining the concept and method of organic synthesis with biocatalysis to realize the precise regulation of natural product synthesis process, which is of great significance for the in-depth study of natural product activity and product development.
